Effect of Ssanghwa-tang(Shuanghe-tang ) added to Cervi Cornu Parvum on Bone Density and Bone Biochemical Marker in Ovariectomized Rats

Abstract
Objectives: As the average span of human life extends, more and more people are at risk of osteoporosis that is one of the typical diseases for the aged people. This thesis presents the effects of Ssanghwa-tang(Shuanghe-tang) added to Cervi cornu parvum on bone density, bone biochemical marker, and fetal calvarial cel(FCC) of the Sprague Dawleys(S.D) Rat that have induced osteoporosis. The purpose is to see how Ssanghwa-tang(Shuanghe-tang) added to Cervi cornu parvum improves the osteoporosis symptoms.

Methods: The first experiment is that Sprague Dawleys Rat were dosed with Ssanghwa-tang(Shuanghe-tang) added to Cervi cornu parvum(KS) for 70 days, one time a day. Two different doses were applied high dose(KSH), and low dose(KSL) groups. The results were compared with a comparing group. The second experiment is that the fetal calvarial cells (FCC) obtained from babies inside pregnant Sprague Dawleys Rat were applied with Ssanghwa-tang(Shuanghe-tang) added to Cervi cornu parvum. The FCC from high dosed and low dosed groups were compared with that from a comparing group.

Results:
1. Femur bone density increases significantly in KSH, KSL. Tibia bone density increase significantly in KSH.
2. Femur and tibia bone ash density increase significantly in KSH, KSL.
3. Calcium of bone increases significantly in KSH. Magnesium and phosphorus of bone increases significantly in KSH, KSL.
4. Osteocalcin and ALP of serum decreases in KSH, KSL but insignificantly.
5. IL-6 of serum decreases significantly in KSH, KSL.
6. OH-P of urine increases significantly in KSH.
7. Calcium and magnesium of serum increase significantly in KSH, KSL. Phosphorus of serum decreases significantly in KSH, KSL.
8. GOT, GPT, creatinine don¡¯t change significantly.
9. Cell proliferation and ALP activity of FCC increase significantly in KSH.
10. Protein and collagen synthesis of FCC increase significantly in KSH.

Conclusion: It was found that Ssanghwa-tang(Shuanghe-tang) added to Cervi cornu parvum have a tendency to make significant increases in bone densities by enhancing bone formation and by retarding bone absorption. It was concluded that Ssanghwa-tang(Shuanghe-tang) added to Cervi cornu parvum activates Osteoblast cells effectively.
